1849年、アメリカ、ボルティモア。闇夜を切り裂く悲鳴と共に怪奇的な殺人事件が起こる。密室に残された母娘の死体。母親は絞殺後に首をかき切られ、娘は殺されて煙突の中に宙づりになっていた。現場に駆け付けた若き刑事エメット・フィールズは、この事件が、高名な作家エドガー・アラン・ポーの一世を風靡した小説「モルグ街の殺人」にトリックや殺害方法が、酷似していることに気づく。酒浸りで生活も困窮しているポーは容疑者と目されるが、狭められる捜査の中、新たに第二、第三の殺人事件が起こる。そのすべてがポーの著作を模倣した方法で…。たて続けに起こる連続殺人は、ポーのアリバイを証明することとなり、ポーは容疑者から一転、著作を汚す犯人を追うことを決意する。しかし、謎の殺人鬼はポーの恋人エミリーを誘拐し、ポーに殺人の偉業を新聞に掲載するように「挑戦状」を叩きつけてきたのだ。“死体”に残された殺人鬼からのメッセージ。見逃したトリック。そして、奪われた恋人の行方を指し示す記号。散りばめたられたパズルを完成し、ヤツを捕らえられることができるのは、全てを知り尽くしているポーしかいない。今、ポーと殺人模倣犯との命をかけた頭脳ゲームの火ぶたが切って落とされた。
